I'm going to isolate plasmid from unknown soil bacteria, is there any simple plasmid isolation protocols? Since using a kit is actually my last option, so I'm trying to use manual protocols instead. I already found one but it turns out that it only works best on gram negative bacteria.
You can use the plasmid extraction method described in this article. This is simple and easier and will save your money.

I usually go with plasmid isolation method called 'Alkaline Lysis' when I need to isolate plasmids from E. coli. You can find the protocol in Sambrook's Molecular Cloning: A Laboratory Manual (you can find the pdf somewhere online I think or visit the web address I added below ). It's the simplest method (other than using kit) and I find it very convenient, among any other plasmid isolation methods. Hope it helps!
