I want to immobilize Nitrile hydatase which is in isolated form to convert Nitrile to Amide.
Where do you want to immobilize it? Need to know the groups on that surface.

General immobilization procedures are described in Hermanson's book (Bioconjugate techniques)
Mukut, easiest way is aminoreactive beads (e.g. NHS activated sepharose (=beaded cross-linked agarose), CNBr activated sepharose), like one would do for affinity chromatography. Alternatively, you may start with carboxy functionalized beads and activate them yourself (NHS/EDC chemistry in MES buffer pH 5.5, protocols usually come with the beads).
