My Sh-RNA plasmid construct kit is having 4 different target sequences against my gene of interest so for knockdown of my gene of interest should I mix all of them before going to start transfection other wise should i try with indidvidual constructs.

Origene technical support team has suggested me to do first individually out of four any two will give me 70 KD if not thye will replace the kit like that they said. After performing individual construct i can mix up them to increase the KD efficiency.

It maybe better to try individual construct to KD. If also not work well. Maybe you can try lentivirus infection instead of transfection. Such as pLKO.1 KD system.
