If the objective is proteomics, PreOmics has a version of their iST kit developed specifically for plant tissue. Given, you do need focused ultrasound (e.g. Bioruptor) for efficient lysis. But a very easy and efficient method for sample prep for proteomics.

Protein extraction from any plant tissue, particularly the woodier plant tissues, is always problematic. The polysaccharide and lignin crosslinked cell walls get in the way both of letting endoproteases to get in to digest the proteins to peptides and to recover the proteins intact from the cells. Then there is the issue of integral membrane proteins, which are almost never recovered unless surfactants are used. The problem with using surfactants is that you can never completely get rid of them in down stream processing.

We partially solved this latter issue in animal tissues,

with the TD buffer system. I've more recently been working on a solution for plants, tested on unicellular algae, but this is still going through the patent process, so is unpublished. Happy to share this with you under an NDA. I'm looking to do more validation in terrestrial plants.
