Different plant extracts will exhibit varying levels of toxicity/effectiveness at different concentrations. So, you will have to use a range of concentration of your plant extract (say ng/ml to mg/ml) to decide the optimal concentration.
For instance, if you desire to check the cytotoxic effect of your plant extract on cancer cells using cytotoxicity assay such as MTT assay, you will have to use a range of concentration and construct a curve by plotting cell survival (%) versus drug concentration (mg/ml) and determine the IC50 value which is the extract concentration that produce 50% cell growth inhibition. IC50 value is correlated with drug potency, i.e. the amount of drug necessary to produce the effect. The lower the IC50 value the more potent is the drug.
