I want to make orthotopic mouse model for colon cancer, but many researchers say, it is very difficult to do that. The problem i am facing is the injection of cancerous cells into the submucosal wall of the caecum, as it is very thin. So i need to get another option for precise injection of cells into the submucosal wall.

It is indeed very difficult, but we are also using injection (HT-29 colon cancer cells) into the caecum wall with euthanazied mice and under microscope.

1.How much volume of cells you inject?
2.You use matrigel or not?
3. what cell count you inject?
4. Do you take support while injecting cells in submucosal layer?

We used two models:
1) mouse CT26. WT colon cancer cells injected in the caecum of Balb/c syngeneic female mice or
2) human HT29 colon cancer cells injected in the caecum of nude / athymic mice.
CT26.WT cells were injected at 1 million / 100 µL with Matrigel.
HT29 cells were injected at 2.5 million / 100 µL with Matrigel.
We did not take support to inject the cells.
