I have used Full strength of MS and Half strength of MS media along with BAP and 2,4 -D , with and without Charcol(.5mg/L/) and for sterilization I used Ethyl alcohol and HgCL2 . But no satisfactory result has come out for last 6 months.As explant I am using spores from mature sorus. But no response yet !
Hi, i ma culture adiantum capillus vernis(sporophyte) in my lab in mixture of soil and some artificial media and get its spores, you can get spores directly from plant fertilize and sterilized.
Spores are available at field but the problem is regeneration. They are not showing any response, no callus in media. So, I wanted to know the media composition and sterilization method. It would be very helpful if you can give me a brief about your methods.
