I am going to isolate the cancer cells from cancerous tissue (colon cancer) to culture and also perform in vitro genetic analysis. Who has experience in this field? Our group have used several protocol but it was not very effective.
If you want to prepare a cell culture, use sterile technique to isolate a specimen of the tissue. I hope you have a laminar flow hood. Mince the tissue with small sterile scissors and put it through repeated trypsinizations to disperse the tissue into single cells. You need to research tissue culture methods and make sure you have the right materials. Since these are transformed (malignant) cells, they may/may not grow in a monolayer, but may prefer to remain suspended in liquid medium instead. "Google" tissue culture methodology and good luck!
