Can anyone suggest any other protocol of mitochondrial ROS generation other than DCFDA assay and JC-1 DYE assay?

We have checked ROS and mitochondrial membrane potentials integrity but need one more experiment to see the effects of our nanoparticles on mitochondrial ROS.

Hi Kunal,

Use MitoSOX Red reagent. The protocol is very simple and you will find with the product sheet.

I do not recommend to use DCF-DA dye for mitochondrial ROS detection. You will find the challenges of using DCF-DA in this article (Kalyanaraman et al., 2012).
