I used to perform germ and Sertoli cell cultures, and I was aware that at one point in the protocol you can get Leydig cells to culture them. You need to decapsulate the testis and treat it with collagenase until the seminiferous tubules are separated. At this point you can take the supernatant that contains Leydig cells. Since you will also have other cell types in here you can separate them in a percoll gradient.
