I am trying to isolate and characterize PGPR strains from rhizospheric soil of paddy fields. In this area there is a high levels of iron content in the soil. My focus is to isolate some heavy-metal resistant PGPR from such soil conditions. But dealing with resource problem, so trying to isolate and characterize them with very basic available resources. Will be very grateful if anyone can suggest me a simple and rapid detection method to isolate and characterize the main or important PGPR strains from such soils, apart from costly metagenomics and conventional biochemical methods which require various media at a good amount.
You can isolate bacteria with PGP function directly from soil dilutions by using selective media such as NFB for N-fixers and NBRIP or Pikovaskya for P-solubilizers, CAS for siderophore producers. For other traits such as production of auxins and ACC deaminase you need to isolate first and then test them.
No Carlos, not yet. Actually it seems very new to me. Just started to working on a PGPR research proposal that I have to submit. I am really a very fresher to research work. So will be very happy if you all can help in that. very less biodiversity related work had been done yet in this region of my country so thought it would be a good idea to work on such area. all the article i have gone through, most of them have preliminary isolation on Jensen's N free media, King'B and NA followed by their biochemical characterization and genetic diversity. But due to less resource and very less work had done in microbiology here most of the media and chemicals are not available on lab, so trying to make it as simple and short as possible.
Do you know if anyone has used metagenomics to study PGPR in fact? I woul dbe interested to find out more about it as we are considering to do this (it does seem like a good idea).
Dear Ayan, Given that the nature of PGPR bacteria is the result of a range of phenotypes - including the expression of antibiotics, competitive agents, motility, etc. - it seems very unlikely that anyone could identify a good candidate PGPR by some genomics approach. You will need to select for a type of PGPR (e.g. pseudomonads, using PSA - CFC plates), then to test them for the appropriate heavey metal resistance, and then to test their ability to colonise soil and plants (using a simple plant-pot system). I would suggest that plate-based selection is realitively cheap - using different media (or supplements in general media) it is possible to determine a range of phenotypes that will let you do some sort of cluster analysis, showing which isolates are more similar than others. From this you can choose a smaller set of representative isolates to test in soils and plants (which takes more time), Regards, Andrew.
