I am trying to see when yeast cells move to G1 (n) after release from nocodazole arrest (2n). I started the experiment by first synchronizing them in G1 (which shows a sharp 1C peak in Flow). Then I released them in YPD for 30min, and then added 15ug/ml nocodazole and incubated them for 30min (which shows a sharp 2C peak). After that I washed them from nocodazole and released them in YPD and tracked them every 20mins. My problem is that I could not see a complete release from nocodazole even after 100min.
