I hope you are aware that rice chromosomes are reported to be very small and cytoplasmic content too being high (Sharma et al 1965). It is difficult to stain somatic chromosomes with conventional stain. Due to the accumulation of silicates. (Fukui1991) the tissues are also very stiff . Kurata and Omura (1978) first applied enzymatic and flame dried method in Rice for demonstrating the clearly shaped rice chromosomes. I think proper pre-treatment followed by EMA method and Giemsa staining is the alternative. It is unfortunate that in India Rice chromosome did not received adequate attention it deserves as important crop.
