In order to check the antimicrobial activity of chemically synthesized nanoparticles, it has to be dissolved. I tried to dissolve the nanoparticles in DMSO (Dimethyl sulfoxide) and methanol, but it didn't dissolve.
The same were loaded in the well during the well diffusion method, and the particles became settled in the well, but only very little activity was encountered.
Please suggest how I can check antimicrobial activity for the nanoparticles.
If you have naked metal nanoparticles, it would not get well dispersed as nanoparticles tend to form aggregates and precipitates. The problem is surpassed by coating the nanoparticles with stabilizers (usually surfactants or polymers). The nature of the stabilizer depends on what kind of solvent do you what the nanoparticles to get dispersed (solubilized) in.

Thank you for your suggestion sir . Since the nano which i have is chemically synthesized, can u suggest what kind of surfactants can be used. Because if it is biologically synthesized nano means the extract itself contains surfactants which forms a cap , but for chemically synthesised i am not sure . So can you please suggest .
sir for colloidal dispersion of nano particles in suitable liquid you can try probe ultra sonication which is more effective than bath sonication, the sonication time we normally carried out for nanofluids preparation is more than 20hrs too but you can try for few hrs is enough. The surfactant mediated stabiliztion is possible but problem is sometimes surfactants will show antimicrobial activity to some class of microbes so you try suspension without surfactant. If sonication method not workout u made small pellets of ur nanopowder and used it for antimicrobial testing. If u alter the ph of solution above or below isoelectric point u can able to suspend to particles colloidally stable but thing is the ph of the solution get altered so the pellet form of nanoparticles if u take for antimicrobial testing gives accurate result.
IF your nanoparticles have been hybrided with polymers, it is easy to dissolve.
I agree with using sonication as a first choice. I want also to suggest a second choice which is trying to control particles surface charge:
You can start by studying surface charge and zeta potential of the particles "you need Zeta analyzer", then you can study the effect of addition of electrolytes "salt" in different concentrations on the surface charge. High value of zeta potential is recommended to get the particles dispersed "high repulsion". Accordingly you can select amount of added electrolyte. Please refer to Martin's Physical Pharmacy and Pharmaceutical Sciences. Clear discussion is found in Chapter of Colloids. Check hydrophobic colloids. Also great discussion is found on chapter of interfacial phenomena about zeta potential.
Addition of surfactant may affect real data about anti-microbeal activity; since surfactants may have anti microbeal activity. If you want to use surfactant; you need a control solution containing same concentration of surfactants.
