I like to use 5-FU as a positive control anti tumor agent in nude mice bearing hepatocellular carcinoma xenograft. Can anyone suggest what dose can I use for the same? Example 20mg/kg/day?
The concentration of a compound for a given xenograft differs significantly based on what cells are used. Even for hepatocellular xenografts, the differing growth curves (http://altogenlabs.com/xenograft-models/liver-cancer-xenograft/) are already enough to warrant different concentrations. That's not even mentioning the route of delivery (injection, oral, etc.) and the location of delivery (subcutaneous, intratumorial, etc.), which also impact the concentration. The previous answers have been around right (5 mg/kg is good) but obviously previous literature and experimentation should determine final concentrations.
