Fatty acids are not soluble in water. Usually the fermentation supernatant has the water and also the fatty acids. It is tedious to quantify the fatty acids in the fermentation supernatant.

For fatty acid analysis, extract your sample with hexane. Then dry into a rotor vapour and again redissolve your sample in hexane. Finally go for GC-FAME analysis or GC-MS analysis for fatty acids.

We do not follow the HPLC for fatty acid analysis. We make a FAME (fatty acid methyl ester) and inject with GC-FID....We receive good results
