I am performing qPCR on a mouse cell line, and I need a recommendation on a control. We typically use IDT Prime Time assays. I assume something like GAPDH would work, but I have never done this on a mouse line before, and I wanted to get someone else's opinion, if anyone has a recommendation.
Unfortunately there is no easy answer. We have used GAPDH- beta-actin and RPL-P0 with success in the past. However, each of these so-called control genes may actually respond to your treatment or conditions. So you must validate that your control gene(s) does not respond. Hi Ian, I used GAPDH as internal control in rat primary culture from cortex in my experiment (and it works). You may start with this and other common candidates as you validate your controls. Good luck.
