I want to measure the total lipid content from Arabidopsis seeds. Please suggest me a simple biochemical assay protocol for the quantification of lipids from dry seeds. Is there any reliable photo-metric measuring protocol available?
Use the soxhlet apparatus with a solvent like petroleum ether or hexane to extract the total lipids. Many resources are available on the web. Or use automatic extraction systems like Soxtherm.
I would suggest grinding the seeds to a fine powder and then weighing the powder. Then do a Bligh-Dyer or Folch extraction and evaporate the solvent and weigh the residue. Soxhlet extraction of the ground seeds with n-Hexane for about 4 hours and evaporating the solvent and recovering the fat portion is a method employed by most researchers for this.
