I have been isolated bacterial strain from Fecal sample... In my preliminary study, those strains are need to be screened based on survival in toxin containing medium during 168 hr incubation.. In research papers, survival study are performed using Phosphate buffer, but not clear mentioned, what kind of phosphate buffers are used at which pH... So can anyone help me to solve these trouble please....

Prepare
1L PBS stock (10X) by adding:
80 g NaCl, 2 g KCl, 14.4 g Na2HPO4, 2.4 g KH2PO4 and make up the volume to IL.
Use 10 times diluted solution of this stock.

Thank you so much sir... Surely I will try this... One more doubt sir , Is there any separate pH range for different incubation period...
Phosphate buffer working pH range is between 5.8 and 8. Generally it is used for pH 7. As Mr. Harish mentioned, that preparation will give pH 7. If you are conducting the same study at different incubation times then maintain the same pH irrespective with the time.
