Can anyone help with problems in detection of P-Met (Y1234 and 1235) without ligand stimulation?

I am not able to detect Y1234/1235 (but basal MET) by western bloting. Increasing first antibody concentration does not help. I do not have a positive control or directed ligands.

It appears that stimulation of hepatocellular carcinoma cells with HGF enhances P-Met (Y1234/5) according to this article (Figure 4 specifically) http://cancerres.aacrjournals.org/content/73/17/5580.full.pdf+html. The efficacy of this treatment depends on the type of cells you are using so first you'd have to determine if they have the proper receptor.
