Can anyone help with Mpp+ cytotoxicity in pc12 cells?

I think first of all check the expiry date of your reagent and if it is not new use an new batch of it.

The second suggestion is to add the concentration of your MPP+ higher than 1 mM to prospect if your MPP is working suitable or not?

Third, there are several methods to assessment the cytotoxicity including MTT, LDH, ROS, evaluating the cell death including apoptosis or necrosis by either florescent microscopy or Flowcytometry, Glucose consumption. I suggest try all of them to confirm the cytotoxicity of MPP on your PC12 cells

Forth, please use a new stock tube of PC12 cells and grow it carefully with love and affection and while you are sure of confluence and healthy of your cells, treat them by MPP. As you know when cell line such as PC12 drives under several passage the morphology and genetics of them could be altered, In the case of PC12 as it is not completely differentiated, maybe in a high passage go for further differentiation and influenced on your treatments,

What was the test that you used to determine the cytotoxicity of your compound? Did your experiment consist of positive control and how was your positive control drug kill the PC12 cells? Positive control should be set by using the drug with the concentration that confirm can kill the cells. It can help you to validate your result.

You should check your PC12 cells if the positive control drug is not working. But if your positive control is working, means your compound may have problem or concentration is too low to kill the cells.
