If you do mma plastic bone for histomorphometry, do you use the same percentage of benzoyl peroxide se in last infiltration and embedding mixture? do you vacuum the bone after embedding?
Do you need it for plain histology or for immunohistochemistry ?
In both cases consider using the Heraeus Kulzer Technovit resins - they are slow curing, several versions are available, they are quite well documented in the literature and the 9100 allows for immunohistochemistry.
The key points of pmma embedding is dehydration in isopropanol and vacuum impregnation (higly recomended to avoid bubbles). For the vacuum impregnation all you need is a desicator jar with vacuum line and a simple memebrane vacuum pump. from my experience it\s worth embedding in simple canisters wich can be sealed with a lid (technovit 9100 should be protected from oxygen while curing)
Thank you for your reply. The two articles I can not access them. I have heard technovit 9100. But have not use it yet. If you personal used this product, please share. I used MMA and it can allow to do limited IHC.
We used an additional gram of dried benzoyl peroxide (per 905mL of solution) in our embedding mixture than our last infiltrating mixture. Using a vacuum to eliminate bubbles is definitely necessary.
