We are screening samples of malarial parasite DNA from human blood samples that we have kept in -20. Using the Kit method, when we check the O.D it comes in the range of 1.6 but when we run it on Gel Electrohoresis their comes no band, while the Ladder shows clear separation. Can anyone please suggest some solution?
No band and no smear? Maybe your sample is degraded. OD shows presence of molecules of DNA but not it condition...
Is their any way out , which can help us as we are having just frozen samples of Blood and we need DNA in proper condition I mean we are ready to change the protocol for isolation, any other protocol for DNA isolation from frozen samples, as we have to do this from those samples only
the protocol attached works well... especially in using stored DNA samples.
Ms Rehman, If you had isolated DNA for PCR based detection of Malarial parasite, I would suggest you to go for nested PCR based detection of Malarial parasite from the isolated DNA; as even degraded DNA element may possess 10-100 target gene sequences which can still be amplified after second round of amplification i.e. at least after 40+40 round of amplification
