I'm currently working with primary cultured hippocampal neurons in which supposed to study localization and trafficking of NMDA receptors, in particular GluN2A and GluN2B.

I have tried following antibodies with no certain luck:

Rabbit polyclonal anti-NMDAR2B Abcam AB65875, 1:200

Mouse polyclonal anti-NMDAR2B BD Bioscience 610416, 1:100

Rabbit polyclonal anti-NMDAR2B Millipore 06-600, 1:200

My work is based on synaptic plasticity I therefore did co-staining of these subunits with with post-synaptic-density protein 95 (PSD95). My PSD95-staining is excellent, and the procedure of immunostaining have been successfully used in our lab for a decade.

Fixation of the neurons are done in 4% PFA for 20 min RT.

Does anyone have experience with immunostaining of NMDA receptors in cultured cells, especially hippocampal neurons? Do you use a specific protocol to identify these receptors, or simply just specific antibodies?
