Can anyone help with cell morphology and passage number?

I am currently doing EMT study. I notice that cells tend to grow in spindle shape after few passages, which are resemble to the EMT-ed cells. What are the possible factors that contribute to this change? Thank you.

What kind of cells are you working with? Primary cell isolates from tissues? If you isolated epithelial cells from a tissue, they can look different over time. What is your media? What kind of serum do you use? Is there TGF-beta in the serum? You need to give us more details.
