I am trying to use an lgl1(Hugl-1) on postnatal mice brain sections. We have had very little luck. Does anyone have an suggestions on antibodies or protocols?
Hi Hannah, I think you are giving too little information. Things can be very different weather you are working with paraffin-embedded sections, cryosections or vibratome sections. IF if notoriously difficult with paraffin-embedded sections, even after antigen retrieval treatments, while IHC can work better, possibly also due to enhancement tricks offered by the detection kits. IF works best on vibratome floating section, but then you'll require a confocal microscope for imaging. Intermediate situation would be cryosections, possibly freezing the brain unfixed in cryoprotecting agents in a mold (Tissue-Tek) and then postfixing after cutting before permeabilizing. I could send you some of the protocols we use if you let me know what type of sections you are working on, but any antibody will require optimization anyway...
