As the Ramus paper explains, Alcian Blue is a cationic dye that interacts with anionic polymers, including both sulfated and carboxylic-acid containg polysaccharides. If your polysaccharide is polyanionic, then it should work the same. If your polysaccharide is not polyanionic, then you need to choose a different method. The periodate-Schiff method is good for most glycans.
