I tried different direct extraction procedures but still getting low signal when detecting at 280 and 320 in a linear gradient of destilled water and methanol with acetic acid , i manage to separate the standars. However i have high signal for anthocyanins which i am not interested in....I am trying to messure Chlorogenic, cafeic, gallic and ferulic acids. Thank for any help!
Unfortunatelly we work with GC method for fatty acids detection
Try to extract with ethyl acetate and water and you will have an enrichment of anthocyanins in the water and the desired acids in the ethyl acetate phase.
You might also try 210nm--gallic acid absorbs well at that wavelength. I'm not sure about the other compounds though.

ACN as a mobil phase and around 320-330 nm gave the best results for chlorogenic acid seperation.

There are many protocol for CLHP of PP
ijf you want analysis only phenolic acid you must useCLHP at 280 nm and not GC
we have working in CLHP of PP there somme publication about PP in CLHP

try...a different mobile phase....I use a binary gradient starting with 2% aqueous phase (2mM natrium acetate+ acetic acid adjusted to pH 3.5) and 98% acetonitrile ; then I vary the gradient up to 30% organic phase; total time 60 minutes for Eclipse C-18 (4.6x150mm, 5um) stationary phase.

Hello. You have to use another mobile phase, e.g., water-acetonitrile.
Have a try with gradient flow (water - acetonitrile), add 0.1% TFA.
If you're doing semi-prep HPLC make sure you add water in to your sample.
This will increase yield and resolution.

you can use colone like (250 x 4,6 mm, taille des particules 5 µm, Macherey 00351179, Nagel, Allemagne) et la pré-colonne CC8 / 4 Nucleodur C18 in non lenaire A = water and formic acide, 95: 5, v / v; and B = / water / formic acide in acétonitrile, 80: 15: 5, v / v / v).
19 min, 3-4% de B ; 19 à 30 min, 4% de B; 30-31 min, 4-6% B; 31-38 min, 6-14% de B; 38-50 min, 14% de B; 50-55 min, 14-30% de B; 55-65 min, 30 à 35% de B; 65-68 min, 35 à 50% de B; 68-70 min, 50-80% de B; 70-75 min, 80% de B; 75-80 min, 80-3% B; 80 à 90 min 3%.
