I have no experience in this field and I really need to know if the results of my back titration are between 30 and 300 TCID50. How can I calculate the TCID50 that I obtained as a result of the back titration? I'm performing a virus neutralization experiment.

In back titration, the working virus in SNT is diluted by serial tenfold
dilutions. The diluted working virus is normally incolulated in 4 to 5 wells per dilution with 50 µL/well which is the volume of virus that is used in the SNT proper. After 2 days, examined for virus cytopathic effect (CPE) and corresponding TCID50 is calculated using Reed and Muench or Spearman Kärber method. You will get virus titre. Take antilog of the titre. If the value of the antilog is between 30 and 300, then it indicates that the amount of the working dilution of virus added to SNT is appropriate and is in range expected for the test.
