I am looking for a p53 inhibitor to use in our in vitro study. Based on the literature, there are three potential candidates I could use: pifithrin-alpha, cyclic pifithrin-alpha and pifithrin-alpha p-nitro (which is claimed as cell permeable). Does anyone have experience with those compounds? Thank you so much.

i suggest to use pifithrin -alpha , in my lab work well in PC3 cells,Bph1 cells and mouse marcophage cells.
Why not use cyclic Pifithrin-α Hydrobromide, which inhibits the transcription of TP53 (human) or Trp53 (mouse)? Needless to say, Pifithrin-μ (=2-Phenylethynesulfonamide) is also useful, which inhibits the binding of p53 to mitochondria, thus preventing bcl-2-dependent apoptosis program.
Thank you, Go J, I am actually trying to figure out what the difference is between Pifithrin alpha and cyclic Pifithrin alpha. It seems they both inhibit transcriptional activity of p53. Do they have different potency or duration? I would like to know if anyone has the first hand information.
