I did antioxidant test using ABTS radial. I get some extreme results need your advice.
- I blended chitosan and cellulose for composites film then test antioxidant of the composite thin film. The unusual results that make me confused.
- Composite from 3-20% of chitosan. After 40 minutes, The antioxidant increase from 19 to 52% for 3-20% of chitosan, consequently. It is however, the unusual thing is cellulose and chitosan thin film have low antioxidant 17 and 28%, after 40 minutes. (I did this test for at lest 4 time, the results still happened).
Can anyone explain why cellulose and chitosan have low antioxidant, but after combine them the antioxidant become high?
Any advice?

Thank you very much. I use nanocellulose fiber. The composition of chitosan I have mentioned above 3 to 20%. It is cellulose nanofiber antioxidant is 17%, chitosan antioxidant is 28%. All samples in the thin film.

For Chitosan it is well-described, including concentration dependence:

The combination will make that the value is the result of adding up the contribution of both which is higher (very simplistically 17+28 makes 45%, this of course too simplistic but together they will have a higher value).
However something else might be happening too, as is also suggested in another mixture/composite:

it could be that apparently the addition of cellulose enables the chitosan to move faster through your sample and therefor give rise to higher values.
Possibly a closer look at the concentration dependence of the various variables in your sample might shed some light on this.

We do think about the combination of cellulose and chitosan lead to easier release of chitosan antioxidant activity and higher antioxidant activity, but we looking for some evident.

Cellulose and chitosan are polysaccharides and most of the reducing ends are not exposed. While preparing composite more reducing ends from both the polysaccharides may open. It may lead to increase of antioxidant activity.
