I am currently trying to apply PMA treatment in THP-1 cells to enable differentiation to macrophage-like cells but I am not sure about their morphology.
Could anyone with more expertise help me?
Ps: I used 5, 10, 20 and 25 ng of PMA in 3x10^5 cells for 48h. The picture is a 100x increase so I could show the clumps of cells formed.

Each clump represents a small cell colony. Cell morphology cannot be judged at the provided magnification. However, the provided picture shows adequate cell adherence, which is a read-out for successful differentiation. In our hands, incubation of THP-1 cells with 10 ng/ml PMA results in successful macrophage differentiation within 48 h.
the macrophages derived cells are adherent cells and branched which are easily differentiated from not differentiated cells (non adherent cells), i was using 50ng/ml for 1x10^5/ml cells for 48 hr
