I don't work with monocytes, but we had good success using electroporation on Primary and CD4+ T cell lines for reporter assays. Lipofectamine 2000 did not work well for me with suspension cells (i.e. T cells). However, in my hands lipofectamine and calcium-phosphate method work extremely well for adherent cells like 293T cells.
I worked with differentiated THP-1 cells. If you want to use lipofectamin 2000 or any other such transfection reagents. You need to used different plasmid and reagent ratio to transfect fix no. of cells in preliminary experiment. After that you need to select best one ratio where you will get best transfection results. If you want to work with electroporation, Experience Lonza's Transfection reagents and electroporator work very best. They have special optimised protocol of different cell line.
