i am dealing with callus generation of citrus limon as the leaves and shoots are very hard i have tried with cotyledons but the results are not up tp the mark...kindly help me out if any one can.

Which is the most actively dividing tissue in the nursery staged plant? You can select a few of them for explant inoculation. Sterilisation could be very challenging. Good luck.
thank you for the response but i have tried it with cotyledons and i want the callus on large scale which is not possible by using such a small explant which is not easily available.
can you suggest me some more ideas which i can implement?
