I am working on a study of agronomic performance of tomato cultivars and I want to see the leaf area variation. Since there are many cultivars that I am working with, I am looking for a simple method which is quick to estimate the leaf area of those cultivars.

Depending on your objectives, instead of leaf area, you may want to measure leaf area index using a ceptometer. Leaf area index will allow you to compare differences in vegetative growth among cultivars.

We manufacture a leaf area meter, the CI-202 that has been used to measure variation in tomato leaves non-destructively. This can be used to quickly measure many individual leaves while they are still attached to the plant.

I'm attempting to do the same thing with Arabidopsis, and have had some success with a plugin called Rosette Tracker. There are other plugins for ImageJ which will do this, although they are based around the leaves being arranged in a non-overlapping fashion, as is nearly the case with Arabidopsis. My method involves first increasing the contrast of the Image using an image processing app, then feeding the resulting images into ImageJ (the version referred to as Fiji):
Open a TIFF of your image, containing a ruler in Fiji and calculate size of pixels in mm
Open Rosette Tracker in Fiji
In Settings, change Calibration to number of plants and rows; set Output; show segmentation
Manually set the scale to that calculated above
In Day vs Night, use Calibrate button to select a saturated region in the centre of a leaf
Set Remove Holes to 1 and Size of Clutter to 2
Run the software and verify that each plant is individually selected in the output window
Note down how on a grid how the numbers correspond to the location of the plants
Click Accept and save your results
