I want to quantify the number of filaments in an actin bundle from a confocal image. How would I plot a graph from it? I want to include Interquartile range and the Man whitney test.
Hi! I assume that you will not use conventional confocal imaging, otherwise the lateral resolution may be not sufficient to reliably distinguish individual actin fibers in a fiber bundle. With sophisticated methods like TIRF or superresolution this may be possible and a density plot (e.g. in imageJmay show peak signals corresponding to individual filaments. For negative controls you may disrupt actin polymerization with certain toxins (i am not shure, but taxol may be suitable)It is quite possible that you may require a transmission electron microscope to perform this task.
