I am working on toxic side effects of some of the commonly used anticancer drugs in chemotherapy regimen in normal cells.
With regard to the same, I have found that a protocol given by Friereich E.J. et al(1966) is followed for converting BSA dose to body weight dose.
But I need to know, practically, how to calculate by using the protocol of Freireich with an example.
Can anyone help me out with this regard.

There are two conversions to perform. I think you should first convert the dose in Body surface area (BSA) in humans to body weight dose. ie: 3700 mg/sq.m (3700/37) is equivalent to 100 mg/kg for humans. Divide this value by 12 (the conversion factor from humans to mice) that is 100/12. this will give you the body weight dose corresponding to mice=8.33 mg/kg. Freireich, EJ, et al. You can follow the calculations presented in the attached FDA guideline. For instance, in the Table 3 of the guideline you will find the interconversion factors among species to convert doses in mg/kg units to doses in mg/m2 units. A different factor applies for each species.

i think to convert human dose to mice dose, it should be multiply by 12.3 instead for deviding by 12.
Cancer drugs are evaluated in dose escalating studies in experimental animals.
Freireich and coworkers concluded on studies in mice, rats, hamsters, dogs, monkeys, the same just acceptable toxicity causing drug dose is found in all species if the drug is expressed in meter square body surface area, A surface is a 2 D unit. It is physiologically not possible to express a drug unit in 2D. wat is needed is a 3 D unit, such as bodyweight or total blood volume. Also Area Under the Curve analyses are 2 D and not helpful in determining/ predicting Drug toxicity.
