You should have standard for the constitute to which you r targeting. HPLC is stabilised half an hour. The standard is injected to HPLC the system will move to gaseous phase and will give graph means at perticular time peak will be recorded and it is the time to inject our sample extract and at specific retention time graph will be excerted and confirms the target constituents
