I am trying to make calcium alginate beads for cell immobilization, sodium alginate solution was prepared in the percentage of 2, 3 and 4 with worm d. water. The sodium alginate solution was dropped in different molar concentration of calcium chloride solution (0.05, 0.1, 0.2 0.3 M). To allow the beads 1 hour to harden, but the beads are not harden. Kindly help me

I used the following procedure in master students thesis preparation and its "worked" well. You can use it if you decide.
Reagents: baker's yeast; NaCl - 0.6%; Sodium alginate; CaCl2 (0.2M Mw 111.0)
Procedure: Disilve 1 g sodium alginate in 15 mL 0.6% NaCl (or saline solution 0.9%) with constant stiring and heating. In beaker make a suspension with 5 mL saline and 1 g yeast. Mix the suspension and sodium alginate solution (1:0.5 ratio). The resulting suspension is transferred to a syringe and slowly added dropwise in cold CaCl2 (50 mL) with constant stiring. After formation of alginate beads stiring continue for 10-15 min. Filtrate and wash well with saline. Stored in a refrigerator with saline solution.
