Hi all. We are conducting a metabarcoding study and need to prepare a phosphate buffer to be used as a first extraction step before using a commercial kit for the rest of the extraction. Our primary issue is time related as molecular grade chemicals will take too long to ship to South Africa and are also significantly more expensive. Any advice would be appreciated.

As long as you can achieve the correct pH and that you can elaborate the buffer is stable, I do not see any wrong with it. With the salt and acid, looks like you are thinking about a buffer with pH around 7.
