I'm using an Ab to detect a molecule on the membrane of the cells using flow cytometry. To confirm the specificity of the Ab I tested the Ab using 2 cell lines: one cell line has the molecule on the surface and one does not express the molecule .

But I'm having a problem because the cell line that does not have this molecule on the cell surface shows signal just as the positive cell line that has the antigen on the membrane, the only difference is that the mean intensity is 1 fold lower compared to the cells that express the protein on the surface.

I reviewed the information of the Ab from R&D and they suggest to

"Stability & Storage:

12 months from date of receipt, 20 to 70 °C as supplied.

1 month, 2 to 8 °C under sterile conditions after reconstitution.

3 months, 20 to 70 °C under sterile conditions after reconstitution."

We got the Ab early last year and has been kept at 4 C after reconsituition without Sodium Azide for more than 1 year, using it several times.

Is it possible that the unspecific signal I'm detecting is due to the fact that the Ab is old or is not properly stroed?
