I am having challenge in determining my alpha cellulose, because any time I measure the alpha cellulose after extraction, the alpha cellulose appears to be higher than the holocellulose. I feel it is wrong or it is possible to make such observation after pretreatment?
The cellulose and all of the hemicelluloses that make up the total polysaccharide fraction of wood, straw, and the like are produced by separating the extractives and the lignin from the original natural material. As lignin, extractives, and hemicellulose were gradually removed, the percentage of the ingredients altered. Alpha, beta, and gamma cellulose are combined to form hollocellulose (HC=Calpha+beta+gamma) if the hemicellulose is fully eliminated. Therefore, it is evident that holocellulose is superior to or on par with alpha cellulose in all circumstances.

Holocellulose should be the total of all your polysaccharides. Alpha cellulose is just the glucose polysaccharide. What method are you using to determine your carbohydrate content? Are you using an internal standard? You may be measuring method losses between samples.
