Potato plant of known variety has distinct physiological trait (resistance to biotic stress of high importance) comparing to other plants of the same cultivar. The trait was inherited in its progeny. Is it enough to claim a new cultivar and pass registration?
if you can demonstrate that the stress resistance is durable ie that the pest/disease will not overcome the resistance, then you have a new fixed phenotype and you may be able to register it. I suggest you contact people involved in plant variety registration and ask them .

Generally the standard is novel, distinct, uniform, and stable (DUS). To me, your variety sounds like it fits that description, as long as the trait was not previously known (in "common knowledge"). Of course, you may have to provide more or less documentation of the new trait, depending on where you choose to register it.

If single trait is unique and has specialty character is enough for registration.
Why not?. What about if the trait is valuable resistance against a disease or a pest?
I think it will be OK with multi-environment experiments to fix the stability of this trait as this is the procedure for new wheat varieties ,,,,,,,
