Hi, I need to calculate a synergy/antagonism between two compounds. One of them, when taken alone, is not toxic, but if you add the non-toxic compound to the toxic one, the mixture causes much greater cell killing than the toxic compound alone.
Software like Compusyn can only deal with two toxic compounds both with nice and smooth killing curve. Compounds with r close to zero confuse it. Suggestions?

There are no strict rules as regarding the toxicity of each compound, actually it depends on the dose. Its properties would also varies according to the phisico-chemical laws. Also, the mechanism of action in special sites or receptors would equally be modified by external selective and exchanfing pressures onward the cell tissues.
On the other hand, if you were more specific regarding the molecules perhaps it would be easier answer your question.
