I am working on determining the binding site of a ligand on bovine serum albumin (BSA). From Job's plot and Scatchard plot analysis I could calculate that the ligand has 4 binding sites on BSA. Now, I would like to determine the binding constants of my ligand for each of its binding sites (4 sites) by fitting my data in the Adair equation. I would like to know the appropriate plotting software where I can fit the data and get the constants or any other manual method to perform this kind of study. Any nonlinear regression software that either contains the equation already or allows you to add it yourself can be used. Some software packages, offer specialized add-ons for enzyme kinetics and/or ligand binding. I suggest you try Grafit, if you can spend some money. A 1-year student edition subscription is $60, and an unlimited single-user academic license is $360. The software is user-friendly for nonlinear regression and allows you to write the equations yourself very easily. It also produces graphs.

You may want to read these classic papers by Gregorio Weber which deal with multiple binding and the Adair equation. They are not easy to get through but are well worth the effort!
