I have made CPNP's and loaded a drug. I have checked drug release pattern in PBS pH 4.5 and 7.4 that states in PBS 4.5 most of the drug is released but in PBS 7.4 shows sustained release drug pattern. My question is that if CPNP's are ruptured in PBS 4.5 earlier (in other words drug is released from CPNP's) then we can suggest that CPNP's are for parenteral drug delivery system but not for orl drug delivery system.

Yes, you are right that there are a no. of factors involving in parenteral route.
I have read a no. of papers regarding CPNP's stating their role in tissue engineering, treatment of diseases etc. How CPNP's play their role in releasing the drug to target and making coating on ruptured bone showing dual effect (1. drug release, 2. Ca coating on bone)

as mentioned above that it is a pH sensitive delivery system with dual modalities of targeted drug release as well as system specific sustained drug release. your system is suitable for the parenteral route for the sustained effects. on the other hand it is also suitable for the immediate release and instant local effects through oral route at the site of low pH (4.5)
Dear Muhammad Usman Munir, in addition to what is suggested in above comments, you should check that if your drug's solubility is controlled by pH. This may be the reason for pH dependent drug release.
I have seen that most of calcium phosphate nanoparticles are large in size (around 400 nm; smallest I have read were 150 nm and largest were over 600 nm). Thus, these nanoparticles will be unsuitable for parenteral administration if larger than 300 nm; they can block fine cappilaries of blood. If this is the case, you should try to optimize your nanoparticles for oral administration e.g. enteric coating or co-administration of an acid-suppressing agent (antacid).

I was also thinking about enteric coating as we do enteric coating in case of aspirin to avoid from the side(gastric ulcer) effect of aspirin.
