Hey everyone,

I want to do Western Blot analyses on the somatosensory cortex of mice. For this purpose, my former colleague used to sacrifice the animals by cervical dislocation and then snap-freeze the brains in methylbutane on dry ice before the brains were stored at -70 degree C. Then the brains were cut on the cryostat in 100um slices and immediately punched while still frozen.

My question is now: Would it also be possible to slice the brains in the cryostat and store the slices (in PBSA f.ex.) first? And then punch them later?

I'd be happy to hear your experiences and opinions!
