I'm performing a RNA-precipitation protocol with specific biotin-TEG labelled probes which bind streptavidin magnetic beads. Over time my enrichments have become significantly lower. My suspicion is the biotinylated probes, which have been freeze-thawed approx 5-6 times by now. The protocol has been altered over time, but not dramatically. Does anybody have a similar experience or could this be due to something else?
I have used biotinylated oligoes a lot, although maybe not with the same "TEG" linkage. I have never noticed the biotin lable being so fragile. If you dissolve your oligo in EDTA containing buffer (like TE), it should not be decoposed during storage. I would rather think about some other issues, like the degradation of RNA in your sample, chage of the procedure, contamination of you biotinylated oligo probe with RNase etc.
