Has anyone tested the Agilent Bioanalyzer RNA chip/assay for running single stranded DNA e.g. by running DNA oligos of known sizes? Can you get accurate readings and therefore interrogate sizes of unknown ssDNA fragments?
DNA chips can be used for ssDNA but the molarities ,also bp and other parameters indicated in the output table will not be exact.
Thanks Jameela. I am hoping someone has run ss oligo standards on either DNA or RNA chip assays and can attest to whether you can calibrate the assay to get exact values for ssDNA. This would be useful for QC during single strand sequencing prep.
the chances for calibration assay for ss molecules would be tough as each time results can vary based on structure confimational polymorhpism.. my bet would be ggo with SSCP and see qc
