Hello, I am trying to screen a few recombinant proteins in a 48-well and 96-well plate format. I need to purify those proteins with Ni-NTA resin but the customised plates are too expensive. Does anyone know a method for Ni-NTA purification in plates or for High-throughput purification? Has anyone customised a purification plate? Or, in care you used customised Ni-NTA plates (from Merk) can you reuse them?

Unofortunatelly i'm not able to provide you hints about the use of 96 well plates since i used it few times in the past but at the end i prefer to use a lower trhouguput approach (growth in 24 deep well and purification in relative 24 single small coloumns)
up to 24 sample in parallel can be easly done by using a small volume gravity flow empty coloumn filled with a FF ni-nta resin and a multichannel high volume pipette to speed up the sample loading and buffer filling

To speed up the washing steps you can also fix the coloumn on a vacum manifold used for DNA (e.g sigma aldrich con VM20-1SET)
